摘要:
目的 探讨三七总皂苷(total saponins of panax notoginseng, PNS)对ApoE基因敲除小鼠动脉粥样硬化的治疗作用。方法 ApoE基因敲除小鼠按照TC水平随机分为6组,对照组给予普通饮食,其它组给予高脂饮食。16周后采集各组血清,分离胸主动脉。油红染色检查斑块; HE染色和TUNEL染色检测心脏左室流出道病变及细胞凋亡; 检测血脂4项和oxLDL-C; 实时荧光定量PCR检测ICAM-1、VCAM-1、iNOS、eNOS、IL-1、IL-6、TNF-α mRNA表达。结果 模型组血清TC、LDL-C、HDL-C、斑块面积比例、左心室流出道内皮细胞凋亡和病理损伤均明显增加(P<0.01),其ICAM-1、VCAM-1、iNOS、TNF-α mRNA表达有增加的趋势,而IL-1、IL-6 mRNA表达明显增加(P<0.01,P<0.05); 与模型组相比,PNS给药后TC、LDL-C、HDL-C有降低的趋势,明显降低了oxLDL-C水平(P<0.01,P<0.05),明显减少左心室流出道内皮细胞凋亡(P<0.05),对斑块面积有减少的趋势,对ICAM-1、VCAM-1、iNOS、TNF-α mRNA表达有降低的趋势,可明显降低IL-1、IL-6 mRNA表达(P<0.01,P<0.05)。结论 PNS对小鼠动脉粥样硬化有良好的治疗作用,可能与调节脂类代谢、保护血管内皮、缓解炎症反应、抑制免疫相关细胞的黏附等有关。
Abstract:
Aim To investigate the therapeutic effects of total saponins of Panax notoginseng(PNS)on atherosclerosis(AS)in ApoE knockout mice. Methods According to TC level, ApoE knockout mice were randomly assigned into six groups. Control group was fed with normal diet, and the other groups were fed with high-fat diet. After 16 weeks, mouse serum and aortas were harvested. The formation of atherosclerotic plaque was analyzed by oil red staining, the proportion of pathological lesion and the apoptosis of endothelial cells in left ventricular outflow tract were analyzed by HE staining and TUNEL. The serum level of lipids profiles, oxLDL-C were detected, and the mRNA expressions of ICAM-1, VCAM-1, iNOS, eNOS,IL-1, IL-6 and TNF-α were detected by qPCR. Results In model group, the serum content of TC, LDL-C and HDL-C significantly increased(P<0.01); the area of atherosclerotic plaque significantly increased(P<0.01); and the apoptosis of endothelial cells and the proportion of pathological lesion of left ventricular outflow tract significantly increased(P<0.01). Also, the mRNA expression of ICAM-1, VCAM-1, iNOS, IL-1, IL-6 and TNF-α in model group increased. Compared with model group, the serum content of TC, LDL-C and HDL-C decreased after administration of PNS. The serum content of oxLDL-C was significantly reduced in PNS groups(P<0.01, P<0.05). The apoptosis of endothelial cells significantly declined as well(P<0.01,P<0.05). The area of atherosclerotic plaque decreased after administration of PNS. The mRNA expression of ICAM-1, VCAM-1, iNOS, IL-1, IL-6 and TNF-α in PNS groups were down-regulated. Conclusions In ApoE-KO mouse model, PNS plays a role in the therapy of AS, which may be due to its modulating lipid metabolism, protecting vascular endothelium, decreasing inflammation and inhibiting adhesion of immune cells.
