摘要:
目的 探讨黑骨藤中分离得到的8个咖啡酰基奎宁酸类单体化合物抑制人类风湿性关节炎成纤维样滑膜细胞(MH7A)增殖的作用,并对其活性较好的2个化合物进行可能的分子机制研究。方法 采用50 μg·L-1 TNF-α活化MH7A细胞。MTS法测定8个单体化合物对MH7A细胞增殖的影响; ELISA法测定细胞上清液中IL-1β、PGE2含量; Griess试剂法测定NO的含量; Western blot检测各组细胞中COX-2、iNOS、NF-κB及MAPK通路相关蛋白表达。结果 与TNF-α模型组比较,8个单体化合物均可不同程度抑制MH7A细胞的增殖和细胞因子的分泌(P<0.05,P<0.01),在200 μmol·L-1浓度下,3-O-咖啡酰基奎宁酸(3-CQA)作用最明显,其次是1,3-O-二咖啡酰基奎宁酸(1,3-DCQA); Western blot结果表明,3-CQA和1,3-DCQA均能明显降低COX-2、iNOS、NF-κB p65、p-IκB的蛋白水平,以及MAPK信号通路相关蛋白ERK、JNK、p38的磷酸化程度(P<0.01)。结论 黑骨藤中咖啡酰基奎宁酸类化合物具有较好的抗类风湿性关节炎活性,其作用机制可能与抑制MAPK信号通路的激活,降低NF-κB活性,抑制下游炎症因子表达,进而减轻炎症反应有关。
Abstract:
Aim To investigate the effect of eight caffeoylquinic acid separated from Periploca forrestii Schltr.on proliferation in human rheumatoid arthritis fibroblast-like synoviocytes(MH7A)and explore the possible mechanism of two compounds with better activities.Methods 50 μg·L-1 TNF-α was used to activate MH7A cells.The inhibition on cell proliferation was detected by MTS assay.The contents of IL-1β and PGE2 were detected by ELISA assay.NO content was determined using Griess reagent.The protein expressions of COX-2, iNOS and the related proteins of NF-κB and MAPK pathway were measured by Western blot.Results Compared with TNF-α model group, the proliferation of MH7A cells could be inhibited by eight monomer compounds(P<0.05, P<0.01), and the contents of cytokines significantly decreased(P<0.05, P<0.01), at 200 μmol·L-1 concentration, the effect of 3-O-caffeoylquinic acid(3-CQA)was most obvious, followed by 1,3-di-O-caffeoylquinic acid(1,3-DCQA).Western blot results showed that 3-CQA and 1,3-DCQA markedly decreased the protein expressions of COX-2, iNOS, NF-κB p65, p-IκB and MAPK pathway including p-ERK, p-JNK, p-p38(P<0.01).Conclusions The caffeoylquinic acid compounds from Periploca forrestii Schltr.have effective anti-rheumatoid arthritis activities, which may be associated with inhibition of the activity of NF-κB/MAPK signaling pathway and reduction of expression of its downstream inflammatory cytokines.
