摘要:
目的 观察迷迭香酸对人肝癌HepG2细胞增殖及凋亡的影响,并探讨Vimentin/NLRP3在此过程中的作用。方法 不同浓度迷迭香酸(25、50、100 μmol·L-1)作用于HepG2细胞, MTT法检测细胞增殖; TUNEL法检测细胞凋亡; Western blot检测相关蛋白的表达水平。结果 迷迭香酸能抑制HepG2细胞的增殖,促进其凋亡,且呈浓度依赖性。与对照组相比,迷迭香酸组能明显上调caspase-3的表达,降低Bcl-2的表达( P<0.05),其作用机制与下调Vimentin/NLRP3信号通路有关。结论 迷迭香酸能抑制HepG2细胞增殖并诱导细胞凋亡,其作用主要是通过下调Vimentin/NLRP3蛋白的表达来实现的。
Abstract:
Aim To investigate the effect of rosmarinic acid on the proliferation and apoptosis of HepG2 cells and the role of Vimentin/NLRP3 in this process.Methods HepG2 cells were treated by different concentrations of rosmarinic acid(25, 50, 100 μmol·L-1).Cell proliferation was assayed by MTT method.Apoptosis was detected by TUNEL, and the expressions of NLRP3 protein and Vimentin were measured by Western blot.The expression of TNF-α and IL-β were investigated by ELISA.Results Compared with control group, rosmarinic acid at 25, 50, 100 μmol·L-1 could inhibit the proliferation and induced apoptosis of HepG2 cells in a dose-depended manner(P<0.05).Rosmarinic acid could increase the the expression levels of caspase-3, but significantly suppressed the of Bcl-2 expression levels(P<0.01).The mechanism might be related to down-regulating the Vimentin/NLRP3 signaling pathway.Conclusions Rosmarinic acid can synergistically inhibit the proliferation of HepG2 cells and induce its apoptosis in a dose-dependent manner by down-regulating the protein level of NLRP3 and Vimentin.
