摘要:
目的 探讨肉苁蓉苯乙醇总苷(phenylethanol glycosides from cistanche tubuiosa,CPhGs)脂质体对大鼠肝星状细胞(HSC-T6)凋亡的影响及其抗肝纤维化的相关分子机制。方法 培养HSC-T6,取生长状态良好的对数生长期细胞用于实验,采用重组大鼠血小板衍生生长因子BB(rrPDGF-BB)刺激HSC-T6,构建体外肝纤维化模型。MTT法测定CPhGs脂质体(29.45、14.72、7.36 mg·L-1)对HSC-T6增殖的影响; AnnexinV-FITC/PI细胞凋亡双染试剂标记后,流式细胞仪检测CPhGs脂质体不同浓度组的细胞凋亡率; 实时荧光定量PCR(qRT-PCR)检测α-SMA、Collagen I、Collagen Ⅲ mRNA的表达水平; Western blot法检测CPhGs脂质体对p-JNK蛋白表达的影响。结果 不同浓度CPhGs脂质体均可抑制 rrPDGF-BB诱导的HSC-T6的增殖; CPhGs脂质体(29.45、14.72 mg·L-1)组凋亡率明显升高; qRT-PCR结果显示,CPhGs脂质体不同浓度组均可下调α-SMA、Collagen I、Collagen Ⅲ mRNA水平的表达,并能降低p-JNK蛋白的表达。结论 CPhGs脂质体能抑制大鼠HSC-T6的增殖并诱导其凋亡,其作用可能与抗肝纤维化有关。
Abstract:
Aim To investigate the effect of phenylethanol glycosides from Cistanche Tubuiosa(CPhGs)liposomes on the apoptosis of rat hepatic stellate cells(HSC-T6)and the molecular mechanism.Methods HSC-T6 was cultured, and the logarithmic growth phase with good growth condition was used in the experiment.rrPDGF-BB was used to stimulate HSC-T6, and the in vitro hepatic fibrosis model was constructed.MTT method was used to determine the proliferation of CPhGs liposomes at 29.45 mg·L-1, 14.72 mg·L-1, and 7.36 mg·L-1 respectively.The apoptosis of CPhGs liposome group was detected by flow cytometry after AnnexinV-FITC/PI cell apoptosis double staining reagent.The real-time fluorescent quantitative PCR(qRT-PCR)was used to detect α-SMA, Collagen I and Collagen III.The expression level of α-SMA, Collagen I and Collagen III mRNA was detected, and that of p-JNK protein in CPhGs liposomes was detected by Western blot.Results Different concentrations of CPhGs liposomes could inhibit the proliferation of HSC-T6 induced by rrPDGF-BB, and the apoptosis rate of 29.45 mg·L-1 and 14.72 mg·L-1 CPhGs liposomes group significantly increased; qRT-PCR results showed that the expression of α-SMA, Collagen I and Collagen III mRNA could be reduced to varying degrees by CPhGs liposomes, and the expression of p-JNK protein could also be down-regulated.Conclusions CPhGs liposomes inhibit the proliferation and induce apoptosis of HSC-T6, and its effect may be related to liver fibrosis.
